Would suggest you talk to him and explain the criticality of the responicibilities.
First talk to his manager and understand how critical is this resource , howmuch time it takes to do the KT .
Get a new replacement for this job( if you get any body internally.) ask the employee to hand over all his responcibilities.after conformation of the manager and the new replacement on proper KT you can release him.
for that at least he should spend 7-14 days(including week ends)
If this does not work out you can go ahead as our frnds suggested above

Reg. Your prob., every employee should follow as per company policy. And you are telling that regarding the notice period even mentioned in his appointment order. So he should obey that one, even though he is in probation period he shouldn't violate that. You just tell him about the policy. First of all check it out abt his resignation date, whether he got acceptance letter of his resignation. Without having all these things he should not relieve as per his own interest. Once he got all the above things, and he want relieve immediately in the sense he should pay one month salary to the company, in the sense we can deduct one month salary frm his F&F settlement. Once he resigned in the sense, from the date of resignation he should not get any leave. So we should not give any leaves to him.

1-You should ask Why He wants to leave the company so early.
If the reason is new job....!!! than fine
2- In probation Notice period should be 7 to 14 days, this is what major companies follows.
3- You can tell him their will be not support for from company for F & F settelment.
do not give releaving letter, hold his last salary. And Master key which effect the candidate is post a letter to candidate residence that if he will not give notice period period than he will be liable to face breach of contract as per appt letter.
